Gingipain inhibition reduced the bacterial load of an established P. gingivalis brain infection, blocked A 1–42 pro-duction, reduced neuroinflammation, and rescued neurons in the hippocampus. These data suggest that gin-gipain inhibitors could be valuable for treating P. gingivalis brain colonization and neurodegeneration in Alzheimer’s disease. However, periodontal disease is a major co-morbidity in Alzheimer’s disease and gingipain inhibitors acting to reduce microglial activation and the expression of pro-inflammatory mediators may well have a significant impact on disease progression. This paper makes a compelling argument to find out.
